Abstract Thymoquinone (TQ), the main constituent of the volatile oil derived from Nigella sativa has shown pharmacological benefits against various diseases while nicotine is an active component in cigarette that is known to be detrimental. This study was conducted to assess the ameliorating effects of TQ on sperm count, membrane, mitochondria and testosterone of nicotine-treated rats. Rats were randomized into four groups: control, nicotine, TQ, and nicotine with TQ. Nicotine (5 mg/kg bwt/day) was subcutaneously injected for 30 days to induce damaging effects on sperm and testosterone level. Rats were force-fed with TQ (5 mg/kg bwt/day) for the following 30 days. Sperm count was reduced in the nicotine group (26.72 ± 1.64 106/mL) but showed a... |

Nicotine is the more abundant component in cigarette smoke. Because nicotine is first metabolized in the liver, our aim was to investígate the effects of nicotine on this organ by biochemical and stereological methods. Male Wistar rats were treated with oral nicotine (ON) diluted in drinking water during 32 days. The control group was treated with drinking water in the same period. Rats were sacrificed 24 hours after last day, the blood was collected and the liver was removed. Lipidogram was performed by enzymatic method and collagen fibers, fat globules and hepatocytes were count in the liver by stereological methods. We observed in control group preserved hepatocytes, with no presence of inflammatory cells. However in the ON group was possible to observe... |

Preclinical studies have shown that repeated stress experiences can result in an increase in the locomotor response to the subsequent administration of drugs of abuse, a phenomenon that has been termed behavioral cross-sensitization. Behavioral sensitization reflects neuroadaptive processes associated with drug addiction and drug-induced psychosis. Although cross-sensitization between stress- and drug-induced locomotor activity has been clearly demonstrated in adult rats, few studies have evaluated this phenomenon in adolescent rats. In the present study, we determined if the simultaneous exposure to stress and nicotine was capable of inducing behavioral sensitization to nicotine in adolescent and adult rats. To this end, adolescent (postnatal day (P)... |

Abstract Nicotine concentration is a key index and directly affects the industrial quality and availability of flue-cured tobacco (FCT). Seedlings transplanted at different times were subjected to different climatic conditions, which were closely correlated with the growth, development, and nicotine synthesis of FCT. An appropriate transplanting time is imperative for ensuring high-quality tobaccos. Hence, a 15N tracing experiment in the field (which included three treatments: FCT seedlings transplanted on 5th, 15th, and 25th May) was carried out to evaluate the influence of different transplanting dates on 15N utilization and nicotine concentration in FCT leaves. Results showed that compared to the tobacco seedlings transplanted on 5th May, the seedlings... |

Nicotine is the most important alkaloid compound in tobacco. One of the major effects of nicotine is stimulation of mesocorticolimbic system. Prefrontal cortex plays a pivotal role in personality and mental state. It is considered the main cause of addiction as it is located in mesocorticolimbic dopamine system. Twenty four male rats were divided into four groups based on nicotine administration dose (0, 0.5, 1 and 1.5 g/kg). After animals were anesthetized, their brains were fixed using transcardiac method. Tissue processing and Golgi staining were performed and the stained tissue sections were analyzed by optic microscope and Motic software. By increasing the dose, nicotine significantly decreased the number of neuronal processes. In the higher dose,... |

In this study, the effect of nicotine on the LDL oxidation by the MPO/H2O2/Cl- system and the effect of HOCl on LDL and some of its components, such as methyl linoleate, vitamin E and the amino acid tryptophan were explored. Nicotine, in micromolar concentrations, enhanced the tryptophan oxidation, either present in LDL or free, in solution. Nicotine also decreased the formation of conjugated dienes and oxygen consumption in a methyl linoleate / HOCl system, and there was evidence to suggest an increase in chlorohydrin formation. Acceleration of the vitamin E oxidation by HOCl was also observed in the presence of nicotine. These data show that the interaction of nicotine and HOCl can promote significant biochemical modifications in LDL particle and some of... |

Nicotine consumption can decrease fertility drive in males through inducing oxidative stress and DNA damage. The color of turmeric is because of a substance called curcumin for which some anti-oxidative and anti-inflammatory properties have been identified. In this study, various doses of curcumin (10, 30 and 60 mg/kg) and curcumin plus nicotine (10, 30 and 60 mg/kg) were administered intraperitoneally to male mice for 28 consequent days and reproductive parameters were determined. The results indicated that nicotine administration (0.5 mg/kg) significantly decreased testosterone level, count and motility of sperms, and testis weight compared to control group. However, increasing the dose of curcumin significantly increased reproductive indices in most of... |

Nicotine is an alkaloid toxic effects of oral cavity. In this study,14 adult Sprague-Dawley rats weighing 230-260 mg (±10 mg) were used as experimental animal. The rats of experimental group (n=6) were nicotinized systemically with nicotine sulphate, 2mg/kg subcutaneously, daily in period of 28 days. Pulp, alveolar bone tissue, periodontal membrane and gingival epithelial junction were investigated in these regions in incisive teeth longitudinal cross-section. Thinning of the collagen fibers in the pulp tissue, vascular congestion, and inflammatory cell infiltration were observed. Mesenchymal tissues that is stained positive for vimentin lay underneath the epithelium. A strong expression of vimentin can be observed in formed periodontal ligament. |

The objective of this study was to evaluate the histopathologic effects of systemic use of nicotine on the rat nasal mucosa. Twelve adult Sprague-Dawley rats weighing 180-220 g, were used as experimental animals. The rats were divided into Nicotine and control groups. The rats of Nicotine groups (n=6) were administered 2mg/kg Nicotine sulphate for 28 days. The rats of control group (n=6) were only administered 1,5 ml physiologic saline solution subcutaneously for 28 days. All animals were sacrified at the end of the study and nasal tissue samples were removed and prepared for histologic examination. The sections were stained with Hematoxylin and Eosin (H-E) and Periodic acid-Schiff (PAS) and Trichrome-Masson were observed under light microscope. E-cadherin... |
